GLP/EHMT1 is a histone H3K9 methyltransferase which undergoes monomethylation to direct chromatin modifying activities and alter gene regulation. This model aims to walk through some of the key features of the enzyme and the various components contributing to the methylation of the H3K9 tail. The model begins with an overview of the electrostatic potentials contributing to site recognition, followed by details of the I-SET and Post-SET domains, site specificity, catalysis and the potential application of inhibitors. This model was adapted from the PDB file 3HNA and its paper (DOI:https://doi.org/10.1371/journal.pone.0008570). H3K9 tail = purple, substrate lysine = yellow, I-SET domain = cyan, arginine = red, Post-SET domain = dark blue, Zn = grey, yellow dotted lines = h-bonds. Important residues are coloured by elements.
